Principles of Fluorescence Spectroscopy
In the field of analysis, fluorescent filters can be used for real-time observation, qualitative identification, and quantitative analysis of biochemical activity. Fluorescence spectroscopy technology has been widely used in the fields of biology and analysis due to its high sensitivity, good specificity, and easy operation.

Most fluorescent instruments, including most fluorescence microscopes, are typically of the fluorescent filter type. A typical system consists of three filters: excitation filter (excitation), dual color beam splitter (beam splitter), and radiation filter (radiation) Excitation filters, usually bandwidth filters, are used to pass fluorescence absorption spectra. A dichroic beam mirror is an edge filter typically used at 45 degrees Celsius; Angle assembly is used for reflecting excitation light and transmitting radiation light. The emission filter is usually a bandwidth filter, and only radiation light can pass through the filter to filter out other spectra, including excitation light.

When the tested sample has multiple fluorescence spectra, narrowband filters are the preferred solution. Narrowband filters only allow the strongest radiated fluorescence spectra, reducing spontaneous fluorescence noise and interaction effects, thereby improving the signal-to-noise ratio of the system.

Application fields of fluorescent filters
Biochemical and quantitative fluorescence detection of nitrogen in tissues and cells, including flow cytometry, fluorescence microscopy, multi-channel real-time fluorescence quantitative PCR, DNA analyzer, blood cell analyzer, hemoglobin analyzer, etc. Cell membrane analysis, immune and molecular diagnostic instruments, single-molecule structure analysis, fluorescence active cell imaging, microstructure and single-molecule imaging, single-molecule protein dynamics on cell membrane, calcium ion detection, drug tracking, and cell structure imaging.

How to choose a fluorescent filter
The principle of selecting a fluorescent filter is to allow fluorescence, i.e. emitted light, to pass through the imaging end as much as possible, while blocking excitation light to the greatest extent possible, in order to achieve the best signal-to-noise ratio. Especially in total internal reflection and multiphoton microscopy applications, slight noise interference affects the imaging effect, so the control of signal-to-noise ratio is quite strict. Usually when we look at the parameters of a filter, we usually look at the transmission spectrum, but the fluorescence intensity is relatively weak compared to the excitation light. It is more necessary to choose a filter based on the cut-off depth. The OD value of the cut-off depth is an important criterion when selecting the excitation filter and the emission filter. Generally, it is required that the 0d value of the intersection point be greater than 5 and the laser excitation be greater than 0d6 in order to obtain a good fluorescence signal-to-noise ratio.

Fluorescent filter series
The fluorescent filters produced by Ruicheng Optoelectronics include single bandpass, dual bandpass, and triple bandpass filters. It has the characteristics of low spontaneous fluorescence, high cut-off depth, small bandwidth, and good transmission. After testing by a large number of fluorescent instrument manufacturers and customers, it fully meets the standards.
